What's the best and quickest way to find out if a Mac has a master password set?

Click on the apple icon in the upper left. Then select:
    System preferences
    security
    filevault

The window will say whether a master password has been set, and if not it will allow you to set one.

As an alternative, use Spotlight (the little magnifying glass at the top right of your screen) and search for 'master password'.
