Defrag in Safe Mode?

    Is it better to run disk defragmenter in Safe Mode.

    Yes. It's better because fewer files are in use by the Windows system during the defrag, therefore a more thorough defrag can be performed. It's better, but it's not a big deal.

    It matters little in XP but it is near mandatory in Win9x.

    True, if you are running win9x it will be a lot less of a pain in the butt in safe mode.
